Gomori trichrome, 16oz.

One step stain (Gomori method) is used to distinguish between collagen fibers and smooth muscle in tissue sections. Bouin’s fixative is generally used as a mordant with this process. The Gomori Trichrome method provided a shorter staining process to Masson’s Trichrome method, yielding similar results. The Gomori Trichrome demonstrates the collagen fibers in a brilliant green shade. This color option is unique to the Gomori Trichrome method.

Fixation:
10% Neutral Buffered Formalin fixed tissue is recommended. Sections should be cut at 5μ.

Procedure:
1. Bouin’s Fixative for 1 hour at 56°C.
2. Wash well in DI water to remove all of the yellow color from the section.
3. Stain with Wiegert’s Iron Hematoxylin. Mix equal amounts of Solution A and Solution B just prior to staining. Stain for 10 minutes.
4. Wash well in running tap water for 10 minutes to remove excess stain.
5. Stain with Gomori Trichrome Stain for 15 to 20 minutes.
6. Place in 0.5% Acetic Acid OR 0.5% Hydrochloric Acid for 2 minutes as a rinse and to differentiate the stain.
7. Dehydrate through 95% ethanol, 100% ethanol and into xylene.
8. Coverslip with Acrylic Mounting Medium or other synthetic resin.

Results:
Nuclei - Black
Cytoplasm, muscle fibers, erythrocytes - Red
Collagen - Green
