Auramine O Stain Kit: Auramine O, Decolorizer, Potassium Permanganate

Our Auramine O stain kit is for the staining of bacteria from cultures or patient specimens suspected to contain Mycobacterium tuberculosis. This system test for the presence of fluorochrome-staining bacilli. Mycobacterial cell wall contains characteristic long chain fatty acids called mycolic acid, which bind to fluorochrome dyes. Positive staining reactions are evident by the stained bacteria emitting an intense yellow or golden light against a dark background. The potassium permanganate suppresses nonspecific background fluorescence.
SPECIMEN PREPARATION
Staining may be performed on smears prepared from specimens either directly or after concentration. The specimen should be smeared in a thin layer on a clean glass slide and allowed to air dry then heat fix for 2 hours at 65°C.
STAINING PROCEDURE
1. Shake the Auramine-O Rhodamine-B Reagent. Place slides on a staining rack and flood with Auramine-O Rhodamine-B and allow to stand for 20 to 25 minutes. Rinse gently with DI water and drain.
2. Decolorize specimen for 2 to 3 minutes Rinse gently with water.
3. Counterstain with Potassium Permanganate and allow to-stand for 3 to 4 minutes. Rinse gently with water and allow to air dry.
RESULTS
Mycobacteria will exhibit a golden or orange-yellow color when stained with these reagents against a greenish-blue background.
