Copper Sulfate Solution is a quick screening method for determining the hemoglobin of potential blood donors. It is the simplest of methods ensuring that a donor meets the minimum Hb level of 12.5g/dL. Specfic Gravity 1.053@ 25°C (Phillips, Van Slake et. al. Method)

Preparation:
Prepare the copper sulfate solution by allowing it to reach ambient temperature especially if exposed to extreme temperatures in transport. Mix well before use. Transfer to a Coplin Jar filling it completely and allow to-stand several minutes. Keep jar covered when not in use. If the solution is used at a higher temperature(s) the specific gravity will be lower which will result in the collection of units from some donors whose Hb level may be inadequate for donation.
Testing:
Secure a specimen via earlobe puncture and place one droplet into the Coplin Jar. For a finger stick, puncture the finger firmly on the lateral aspect of one of the middle fingers. Wipe away the first drop to prevent excess tissue fluid from giving a false low result. If the donor’s Hb is 12.5 g/dL or greater the droplet will sink to the bottom of the jar either immediately or within 15 seconds . Hb levels which are less than 12.5 g/dL will float (eventually sink over time, several minutes). Typically, the solution should be changed by allowing the maximum of one droplet of blood per mL of Copper Sulfate Solution. It is suggested that when using a Coplin Jar with a filled volume of
50 mLs, approximately 40 specimens can be tested prior to changing.
Quality Control:
Copper sulfate solutions may be quality controlled upon receipt to ensure the correct specific gravity (ideally tested at 25 degrees centigrade). The ideal long-term storage temperature is 15 to 25°C. If the product is stored at this temperature and remains tightly sealed in the unopened 16oz containers received, the specific gravity will not changed over the assigned shelf life.
Disposal:
Follow the regulatory requirements for disposal of this reagent according the to local, State and Federal guidelines set forth. Copper Sulfate Solution is slightly corrosive.
