Alcian Blue (pH 1.0) Stain Kit
Details
The Alcian Blue (pH 1.0) Stain Kit is intended for use in the histological visualization of strongly sulfated mucosubstances.
Strongly Sulfated Mucosubstances: Blue
Nuclei: Red
Background: Pink
Uses/Limitations:
- For In-Vitro Diagnostic use only
- Histological applications
- Do not use past expiration date
- Use caution when handling these reagents
Control Tissue:
- Small Intestine
- Appendix
- Colon
Procedure:
1. Deparaffinize sections if necessary and hydrate to distilled water.
2. Incubate slide in Acetic Acid solution for 3 minutes.
3. Incubate slide in Alcian Blue (pH 1.0) solution for 30 minutes at room temperature or 15 minutes at 37° C.
4. If desired, rinse slide briefly in Acetic Acid solution to remove excess Alcian Blue.
5. Rinse for 2 minutes in running tap water followed by 2 changes of distilled water.
6. Stain slide in Safranin O Solution for 5 minutes.
7. Rinse for 2 minutes in running tap water followed by 2 changes of distilled water.
8. Dehydrate through graded alcohols.
9. Clear, and mount in synthetic resin.
